This is a rest day, aka a day off. Depending on where you're from, you might also call this a holiday. It's basically any day you have off from work or school.
This is a jukugo word, which usually means on'yomi readings from the kanji. You know the on'yomi for 休, but since 日 has two on'yomi readings, here's a mnemonic to help you remember which one to use:
On every day off, you eat not one, but two 休-cumbers. Gee, two (じつ)? Take it easy!
休日はワインをのみます。
I drink wine on holidays.
今日は休日なので、ピクニックにきました。
Today is a holiday, so I came for a picnic.
休日にメールしてこないで下さい。
